Development of a biosensor assessing SARS-CoV-2 main protease proteolytic activity in living cells for antiviral drugs screening

Dear Editor, The Coronavirus disease 2019(COVID-19)pandemic is caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).The genome of this virus encodes two overlapping polyproteins,pp1 a and pp1ab(Wu et al.,2020;Zhou et al.,2020).The functional polypeptides are released from those two polyproteins by extensive proteolytic events,mostly mediated by main protease(Mpro,also known as 3CLpro).It cleaves the polyproteins at no less than 11 conserved sites to produce non-structure proteins(Nsps)(Wu et al.,2020),and no human protease has been found to share similar cleavage specificity(Wu et al.,2020).Since these cleavage events play an essential role in viral replication and pathogenesis,Mpro is considered as one of the most attractive targets for drugs development to combat the ongoing COVID-19 pandemic(Pillaiyar et al.,2016).
