Solid-phase alkylation:a low-loss and anti-interference sample preparation strategy for low-input proteome profiling

Deep proteome profiling from trace biological samples are needed for important biomedical applications[1],but the sample losses from traditional sample preparation procedures are a major limitation[2].Therefore,considerable efforts have been made to improve it[3,4].Filter-aided sample preparation(FASP)developed by Mann's group[5]is recognized one of the most popular meth-ods,by which protein extraction,reduction,alkylation,and diges-tion are performed on a commercial spin filter.However,the strong detergents that are often utilized to aid cell disruption and hydrophobic protein solubilization are often incompatible with proteolysis and mass spectrometry(MS)analysis;they neces-sitate removal using multiple ultrafiltration operations,which not only increase sample handling time but also lead to sample loss.Recently,to improve proteome sample preparation,in-situ sample processing techniques such as nanodroplet processing platform(nanoPOTS)[6]have been developed to improve the efficiency and recovery of sample preparation through reducing the surface loss and enhancing the collision possibility of reactive molecules in a micro-scale confinement space,which enable the preparation of sub-microgram starting materials.
