Retina cell atlases of multiple species and an online platform for retina cell-type markers

The retina is a complex visual tissue that perceives light signals because of an exquisite orchestration of high cellular complexity(Fig.1A).It is of great significance to explore the transcriptome diver-sity of each cell type for comprehensive understanding(Voigt et al.,2020b).Unlike conventional sequencing methods,single-cell RNA-sequencing(scRNA-seq)and single-nucleus RNA-sequencing(snRNA-seq)accurately profile transcriptomic characterization of in-dividual whole cells and nuclei,respectively.Especially,closely related neuronal cell types can be similarly discriminated by these two sequencing methods(Bakken et al.,2018;Hedlund and Deng,2018).The application of scRNA-seq has produced retina datasets of several species,including human(Voigt et al.,2020a),macaque(Peng et al.,2019),mouse(Hoang et al.,2020),chick(Yamagata et al.,2021),Xenopus,and zebrafish(Hoang et al.,2020).However,establishing a comprehensive understanding of retinas across multi-ple species is challenging due to cellular heterogeneity(Peng et al.,2019).The lack of a multispecies retinal single-cell dataset platform impedes the identification of orthologous cell types across species.Here,we apply snRNA-seq technology to profile the retina cellular heterogeneity of four species that were not previously reported,as well as establish a dedicated platform for retina single-cell atlases,including ten species.
